Rashford missed the start of Gareth Southgate’s training camp due to “personal matters”.
Kalvin Phillips also delayed joining up for the same reason but is expected back soon.
The FA confirmed the pair’s delay at the same time that Jude Bellingham and Levi Colwill pulled out of the squad, taking the number of withdrawals from the original squad to five.
Boss Southgate had already been hit with a trio of withdrawals.
James Maddison, Lewis Dunk and Callum Wilson all miss out due to injury.
Manchester City star Rico Lewis has been called up to the Three Lions squad for the first time, as has his former team-mate Cole Palmer following his impressive start to life at Chelsea.
Aston Villa centre-back Ezri Konsa could also win his first England cap after earning his first call-up.
Rashford returned to the United starting line-up in a 1-0 win over Luton at the weekend, having missed the Fulham victory because of injury.
The forward has just one Premier League goal this season but Southgate has backed him to start scoring again.
The England boss said: “In the end — and respectfully — with the big bucks, comes the big pressure.
“He is the guy that got all the goals last year.
“I’m sure even Harry is not thinking I’ve got to be the one to score.
“Maybe with Hojlund scoring now at United, that will help but you know over a period of time Marcus has scored goals so that’s going to happen again.
“It’s just a case of getting a couple and then he’ll be off and running.”
